Skeletal stem cells are residents of the skeletal system and engines for skeletal development, homeostasis, and repair.  The fine balance between their renewal and their multipotency is dynamically controlled at multiple levels to adapt to the needs of various situations. Dysregulation of skeletal stem cells and their descendants is a major cause of skeletal disease, such as osteoporosis, osteoarthritis, and skeletal dysplasia.  Inspired by genetic studies of human skeletal disorders, the Yang lab at the Van Andel Institute identified several new molecular mechanisms that control sumoylation and heterochromatin maintenance important for the fate, and homeostasis of skeletal stem cells during skeletal development, aging, and disease.
